It looks like that setting was removed with iOS4, but here is something you may want to try in lieu of jailbreaking or buying an alternate browser that might allow that option:

Make a bookmark for the url that you wish to use as a homepage. Then move that bookmark to the top of the bookmark list

once you open Safari browser it will be the top bookmark and one click you can access that url or page as your opening page
